High Performance Liquid Chromatography or HPLC
, is the process of purifying or isolating individual compounds from a mixture of multiple compounds. This process is used in biochemistry and analytical chemistry to study each component in biological and chemical materials.
This chromatography technique was pioneered by Mikhail S. Tsweet, a Russian botanist. He started with a simple experiment to try to separate the pigments of leafs. His first experiment used a solvent (extract of plant leaves) that was passed through a column of particles. He noticed that as the solvent passed down the particles, it separated into rows of different colors. The conclusion was that these different rows of colors were basically different compounds from the plant solvent.
This technique was further advanced with HPLC. Research showed that pressure was responsible for separating the compounds. As technology advanced and instruments were able to create more pressure, even the smallest compounds could be identified. With the use of improved injectors, detectors, and columns, more accurate measurements and results were possible.
HPLC uses a column, detector, computer (with chromatography software), solvent, pump, and an injector (also called sampler or autosampler) to apply the sample. The injector delivers the sample into the column. The pump is then used to create pressure and combine the solvent with the sample. The solvent will then separate the components of the sample and be sent to the detector. The detector can identify each component and send the desired component to waste for collection. The computer will manage the pump, column, and record data.
High Performance Liquid Chromatography has many uses. The most popular use is in the pharmaceutical industry. The use of HPLC allows pharmaceutical companies to determine the content of the active ingredient in medication in its purest form.
Companies also use HPLC to study the stability of pharmaceutical products, figure out how much is required to be effective for patients, and find what is the right dosage to use in their products. Of course, HPLC also has its uses on analyzing various compounds for the advancement of science. By isolating compounds from original compounds, we can study the origin, nature, and effect of the original compound.
As effective as HPLC is, a new method has been developed called UPLC. UPLC is a method that helps scientists analyze more samples, faster, and with more accuracy. The process takes from HPLC and uses re-engineered tools from column design, injectors, pumps, and detectors to produce a better result.
