A number of antibodies are specific to CD44. A transmembrane cell-surface glycoprotein, it is expressed on a number of cell types including B cells, macrophages, monocytes, thymocytes and peripheral T cells. Its main function is in cell adhesion, although it is also involved in cell-to-cell interactions and cell migrations.

CD44 features in a number of cellular processes, including hematopoiesis, lymphocyte homing and leukocyte activation. It is also expressed in the metastasis and development of certain tumours. Antibody suppliers, such as us at Novus Biologicals, provide CD44 reagents for cell surface marker studies, as CD44 is expressed in some breast and prostate cancer stem cell lines.

Foxp3 is a highly conserved transcriptional regulator of the forkhead/winged-helix protein family, and is essential for homeostasis in the immune system. A marker protein, it is specifically expressed at high concentrations in CD4+CD25+ regulatory T cells, where it has a regulatory role and can help suppress T cell proliferation. Although CD44 is known to be expressed in regulatory T-cells, its role is less clear. However, in 2009 Liu et al demonstrated a link between Foxp3 and CD44 expression.

The research involved analysis of Foxp3 levels in CD44 positive T regulatory (Treg) cells, using a variety of assay techniques. IL10 antibodies were also used to determine levels of the immunoregulatory cytokine interleukin 10 in a variety of thymocyte subsets. Suppressor activity in populations of spleen Treg cells was also investigated.


The results showed two distinct subsets of Treg cells in the thymus, each with specific staining features. Multi-assay studies at cellular and molecular levels indicated a positive correlation between CD44 and Foxp3 expression in thymocytes. Additionally, it was shown the suppressive effect of Treg cells on T cell production could be blocked by anti-IL10 antibodies. The study showed that CD44 has functional relevance in determining Treg subsets, which are modulated by IL10 and Foxp3.
