Comparative Analysis of Plasmodium Vivax Plastid Genome
Plasmodium vivax is responsible for causing greater than 70% of human malaria cases in southeastern asia and the Indian subcontinent. The rising severity of the disease and the resistance shown by the parasite towards usual therapeutic regimen has put forth a demand for a novel drug target to combat this disease.A3-4 membrane layered organelle called Apicoplast and its genome are being looked upon as a potential drug target. This organelle has been reported among all Apicomplexans including all Plasmodium species and is considered indispensable for the survival of the parasite. Apiccoplast is prokaryotic in origin harboring a circular genome with genes from blue and red algal lineage, similar to chloroplast DNA.The genome is believed to be conserved among all Apicomplexans. Through the genes of Apicomplexans. Through the genes of Apicoplast genome are believed to be of major functional importance, the proteins encoded by these genses are not characterized as yet. except for a few reports, this genome has not been detailed from one of the major human parasite Plasmodium Vivax.Through the genes of Apicoplast genome are believed to be of major functional importance, the proteins encoded by these genses are not characterized as yet. except for a few reports, this genome has not been detailed from one of the major human parasite Plasmodium Vivax.
we are the first group to report any complete gene from this genome of P.vivax. we have also characterized other major genes of this genome, including large subunit ribosomal RNA gene. sufB,clpC,RNA Polymerase C genes from other plasmodium speciese showed about 8-13% differences at both nucleotide and amino acid level .the Apicoplast was also colocalized in P.vivax infected blood smear obtained from the field, using antibodies against EFTUA peptides raised in Swiss albino mice.we are the first group to report any complete gene from this genome of P.vivax. we have also characterized other major genes of this genome, including large subunit ribosomal RNA gene. sufB,clpC,RNA Polymerase C genes from other plasmodium speciese showed about 8-13% differences at both nucleotide and amino acid level .the Apicoplast was also colocalized in P.vivax infected blood smear obtained from the field, using antibodies against EFTUA peptides raised in Swiss albino mice.
