First, how do you separate yourself from the multitude of competition you have?
Networking. If you have a referral or you know someone within a company you have a great advantage. Often times that person can be a great reference and a way to get your resume to the "top of the pile". The only way to get a job is to get an interview, and networking is a great way to get one.
Have knowledge on the company you're applying for. When a potential employer asks why you are interested in working for them, it's good to have a sincere and valid understanding of the job you'll be taking and how you can benefit the company in the future.
Be aware of your skills and use them to your advantage. Instead of trying to learn new skills to find a job, look for a job that needs the skills you already have and are proficient at. If you have customer service experience, search for a job that is mainly customer service. After you've exhausted job leads that you already have the skills for, branch out and look for other opportunities in fields where you might not be as proficient.
Play the numbers game. You obviously have a preference of which job you want, but the more resumes you send it, the higher chance of hearing back from an employer.
A good way of raising your chances of an interview is to make different resumes. Since you'll likely be applying for different positions, design two or three resumes that highlight and focus on different aspects of the skills and experience you can bring.
Be confident. When you start and interview and you're looking at your future boss' face, try your hardest to remain calm. There's no reason to be afraid or intimidated.
