ABSTRACT

Human immune virus type 1 (HIV-1) is the cause of AIDS disease and is a member of retroviridae. The genomic structure of this virus is almost similar to the other members of this family and consists of gag, pol, env genes variety of the host cells which represent the CD4 receptors on their cellular surface are the victims of this virus.

As most of these cells are involved in immune response of our body, the destruction and termination of them will cause deficiency in immune system and ends in death of the host due to different kinds of infections.

Now a day this virus is one of the major reasons of death among different kinds of diseases in the world.

In this study we have tried to establish the quick, sensitive, and new technique of VISUAL DNA CHIP for detection of HIV-1 infection.

For this reason, first two pairs of primers were designed for gag gene of the virus, then the RT Nested PCR was done for these primers, then by using of dNTP which were labeled by deoxygenize and specific probes the VISUAL DNA CHIP technique was designed for the samples.

Probes were bond covalently on the surface of DNA chip which were hybridized to the products of final step. At the end of this step and washing, DNA chip was subjected to specific anti deoxygenize anti bodies which were mixed by alkaline phosphatase enzyme in the presence of BCIP/NBT substrate. Forming the blue color represents the presence of virus. The advantages to this technique are: Specialty, high sensitivity, short time, easy and doable in most of the labs and also cheaper in comparison with similar techniques.
